Для принятых пяти компоновок самолета были проведены исследования по аэродинамике, устойчивости и управляемости полета. Для каждого варианта груза определялось его наивыгоднейшее размещение на фюзеляже (по длине и высоте), исходя из условий:
- уменьшение лобового сопротивления;
- ограничение разброса моментных характеристик;
- получение нужных центровок;
- ослабление динамического воздействия вихревой пелены, сходящей с грузов, и газовых струй двигателей на хвостовое оперение (бафтинг).


На обоих летающих «Атлантах» в 80-е годы прошлого века было совершено более 150 полетов по доставке на космодром Байконур всех крупногабаритных элементов советского челнока «Буран» и ракеты-носителя «Энергия»
